Try this: DS-2 -> Big Muff -> V2
The secret of his sound: the wah AFTER the distortion/fuzz. I know many people hate this sound of wah so I hope it works for you. John use it in these days in the same way. DS-2 get a buffer to fight the little tone suck of the V2. Try to combine distortion and fuzz to get this really cranked up sound for songs like "Scar Tissue". But danger! The V2 fuck you from behind with the volume boost, look out

I already use the DS2 > Big Muff > Wh10 and I like this combination...but I wanna an advice to obtain a similar sound, for example how to set up the knobs of the pedals...

For the exact sound, you need his equipment
My settings for the typical SA sound are:
V2 = guitar mode, effect depth = 10
Boss DS-2 (Turbo Mode) = level: 3 o'clock, tone: 12 o'clock, distortion: all the way up
Big Muff = volume: 2 o'clock, tone: 1 o'clock, sustain: all the way up
I use a Fender '62 Jag and a Marshall JCM 2000 DSL 401 and with this sets it works best! If you have a Strat or a Tele as a main guitar, use a little more tone, my Jag got so much highs, I have to hold the tone poti back a bit
Try it and you will see...but another equipment - another settings! Try with my idea and you find your/John's sound, I'm sure

I have a Fender Standard Stratocaster Mexican 2009 and a Fender Stage 185 amp with this settings: Bass 6 - Middle 2 - Treble 8 - Reverb 7 - Gain 2 (John uses Gain at 4 but for me it's too distorted. The rest are the JF's settings, I found this in a web site). Do you think it's good?

Don't believe only websites! Try, try and TRY!
For the SA sound I set my Marshall this way:
Bass 5 - Middle 7 - Treble 5 - Gain 5 - Reverb 2
These are the settings for the clean channel, on the crunch channel I set gain on 2.
I got a really good John-sounding sound with this settings and my Jag

Hey dude, the 185 is solid state right?
That can greatly affect how the amp reacts to the pedals. If i were you i would save up some money for a nice tube amp like the DSL401 or a marshall jubilee like me
Anywho, i think you have your eq settings dialled in a little wrong. On my jubilee i run it Bass 10 - Middle 8 - Treble 3 - Presence 4.5. I also turn the master volume to about 6, input gain pretty much cranked full (clean channel) then use my Weber Mass 150 to bring it down to useable volume. This gives me a very awesome frusciante tone.
For pedal settings, Chili_Pat had it pretty much spot on.
But experiment: get a really good frusciante sample, and fiddle with your settings till it pretty much sounds the same. Have fun!

But John run his Jubilee in a very similar way that you a0n09, that must be a Chili sound

I tried the Chili_Pat's settings and I'm very satisfied but I think I'll try the a0n09's settings and I'll choose the best
